Source Pathogens 2020 Aug. Figure 5. N-acetyl-l-cysteine (Abmole Bioscience Inc, Houston, TX, USA)
Method Cell Viability Assay
Cell Lines Caco-2 cells
Concentrations 10 mM/well
Incubation Time 6 h
Results It was shown in Figure 5A that pre-incubation of Caco-2 cells with NAC for 3 h before exposure to trophozoites could block the release of Cyt-c. The activation of CASP3 was diminished by NAC in trophozoites-treated cells
Protocol (for reference only)
Cell Experiment
Cell lines RAW264.7 cells
Preparation method RAW264.7 cells were stimulated with 1st LPS for 12 h followed by incubation with 2.5 mM NAC for 1 h before 0.04 mM metformin and 2nd LPS stimulation. In parallel experiments, RAW264.7 cells were pretreated with 100 μM TBHP for 20 min before 2nd LPS stimulation. The production of O TNF-α and P IL-6 was then determined via ELISA assays.
